ABSTRACT Background: The American cutaneous leishmaniasis (ACL) is expanding in peri-urban environments. Methods: An entomological survey was conducted in the area of the occurrence of an autochthonous urban case of ACL. Sandflies and a parasitological slide of the human case were submitted for molecular diagnosis. Results: Nyssomyia whitmani and Ny. antunesi were the most frequently collected species. Ny. whitmani and Bichromomyia flaviscutellata were positive for Leishmania guyanensis and L. lainsoni, respectively. The human case tested positive for L. lainsoni. Conclusions: Sandflies and Leishmania parasites present in urban forest may occur frequently in nearby domiciliary environments; thus, these areas must be monitored.

ABSTRACT The municipality of Paracambi (Rio de Janeiro, Brazil) reports sporadic cases of American cutaneous leishmaniasis (ACL). Previous studies detected Nyssomyia intermedia (Diptera: Psychodidae) as the main vector in the region, but its spatial distribution and the presence of other vector species have not been evaluated. This study aims at filling this knowledge gap, by studying the ecology of sand flies, their spatiotemporal distribution, and correlation with land use/cover. Two campaigns of monthly sand fly collections using light traps and manual captures were conducted in 1992-1994 and 2001-2003. Females were dissected to detect natural Leishmania infections. The spatial distribution of sand flies was assessed using kernel density maps. Correlations with land use/cover were evaluated by extracting satellite imagery data around the capture points. A total of 17,232 sand flies from 13 species were captured. Medically important species included Ny. intermedia, Migonemyia migonei, Pintomyia fischeri and Ny. whitmani. No Leishmania-infected females were detected. Highest densities were detected in the peri-urban areas Cascata and Sabugo, and in rural areas São José and Mutirão. Ny. intermedia had statistically significant correlations with pasture and agricultural areas. Present results strengthened that Ny. intermedia and Mg. migonei are the main local ACL vectors. Correlations with land use evidence the association between ACL and anthropic environmental change.

ABSTRACT OBJECTIVE To perform an entomological survey, evaluating the circulation of Leishmania spp. in sand flies captured from the new foci of visceral leishmaniasis (VL) in the coastal region of São Paulo state. METHODS Sand flies were captured from November 2016 to September 2018 using light traps of the Centers for Disease Control (CDC), in the neighborhood where VL cases were reported. Leishmania spp. circulation was evaluated by gut dissection and molecular analysis of the females captured. RESULTS Nyssomyia intermedia was the more frequent species (90,7%) within the 1,203 sand flies captured. We found no flagellates in dissected females, but two pools containing females of Ny. intermedia presented DNA of L. infantum. CONCLUSION Our results suggest that Ny. intermedia might be involved in the establishment of this new VL focus in Sao Paulo. However, before incriminating this species as a L. infantum vector, further studies should investigate other vectorial capacity parameters, including competence, survival, and feeding habits.

ABSTRACT

Despite some phlebotomines being well recognised as vectors of leishmaniasis agents, vector importance of those belonging to the genus Trichophoromyia has not been extensively studied. The present study provides evidence regarding the putative vector role played by some species of Trichophoromyia on leishmanine enzootics, based on literature reports and findings obtained from field experiments conducted in the ecotopes of Pará State, Brazil. The species Th. ubiquitalis, Th. velascoi, Th. auraensis, Th. ininii and Th. brachipyga possess minimal criteria to be included in the list of suspected leishmanine vectors. However, knowledge on man-biting behavior, substantiation of vector competence and determination of epidemiological implications are limited for all of the above mentioned species. Published studies together with present data draw attention to prioritize these phlebotomine species in entomological surveillance programs and studies on experimental susceptibility to Leishmania spp. infection.

Abstract Introduction: Sandflies are known for having vector species of the tropical disease Leishmaniasis, a disease which is of an endemic nature in Western Boyacá, where the town of Otanche is one of the main source of Cutaneous Leishmaniasis. Objective: To identify the species of sandfly present in an endemic area of cutaneous leishmaniasis in West Boyacá. Methods: The search and collection of sandflies was carried out using CDC gravid traps, over a period of twelve hours (18:00- 06:00). Identification was carried out by revising the genitalia on both male and female samples under a microscope. The distribution took as reference households with a history of people infected with this disease, locating them intra, peri and extra domicile. Results: Were recollected 361 individuals (252 females and 109 male), belonging to 9 genres and 16 species. 60% of all recollected phlebotominae consists of Nyssomyia yuilli and Nyssomyia trapidoi. Other species recollected and relevant, due to vector precedent, are Lutzomyia hartmanni, Psychodopygus panamensis, Lutzomyia gomezi and Psychodopygus carrerai. Conclusion: It was established that, due to its abundance and vector precedent for the country and the area under study, Nyssomyia yuilli and Nyssomyia trapidoi constitute the species of phlebotominae which may be involved in the transmission of cutaneous Leishmaniasis in the region.


Resumen Introducción: Los flebótomos, son conocidos por tener especies vectoras de la enfermedad tropical Leishmaniasis, enfermedad que se presenta con carácter endémico en el occidente del departamento de Boyacá, donde el municipio de Otanche es uno de los principales focos de leishmaniasis cutánea. Objetivo: Identificar las especies de flebótomos presentes en una zona endémica de leishmaniasis cutánea en el occidente del Boyacá. Métodos: La búsqueda y recolección de los flebótomos se realizó con trampas CDC durante doce horas (18:00- 06:00), tomando como referencia viviendas con antecedentes de personas que hubieran tenido la enfermedad, ubicándolas en el intra, peri y extradomicilio. La identificación se realizó por medio de revisión del órgano genital de machos y hembras al microscopio. Resultados: Se colectaron 361 individuos (252 hembras y 109 machos), pertenecientes a 9 géneros y 16 especies, de las cuales, el 60% de toda la flebótomofauna recolectada está representada por Nyssomyia yuilli y Nyssomyia trapidoi. Otras especies colectadas y con importancia por antecedentes vectoriales son Lutzomyia hartmanni, Psychodopygus panamensis, Lutzomyia gomezi y Psychodopygus carrerai. Conclusión: Se estableció, que por sus altas abundancias y por sus antecedentes vectoriales para el país y para la zona de estudio, Nyssomyia yuilli y Nyssomyia trapidoi, constituyen las especies de flebótomos que pueden estar implicadas en la transmisión de leishmaniasis cutánea en la zona de estudio

ABSTRACT The study aimed to determine the toxicity of lambda-cyhalothrin, alpha-cypermethrin, and deltamethrin in L. longipalpis, through concentration-mortality bioassays. The test here was performed following WHO guidelines, but instead of using exposure WHO recipients and impregnated papers, 250 ml Wheaton glass bottles treated with 1 ml of insecticide solution were used. Batches of ten females of L. longipalpis were exposed to five concentrations of each pyrethroid that caused between 5 and 100 % mortality in this species. After 1 h of exposure, the females were transferred to observation recipients, and mortality was recorded 24 h later. The lethal concentrations (g/ml) that killed 50 and 95 % (LC50 and LC95) of the exposed L. longipalpis females were 0.05 and 0.86 for lambda-cyhalothrin, 0.24 and 3.62 for alpha-cypermethrin and 0.53 and 4.72 for deltamethrin. Based on the LC50 obtained, lambda-cyhalothrin is the most toxic pyrethroid for L. longipalpis, followed by alpha-cypermethrin and deltamethrin. It is expected that these data may be useful in studies on the effects of sub-lethal concentrations of the three pyrethroids on the behavior of L. longipalpis and studies on the vector susceptibility to these pyrethroids.


RESUMEN El objetivo del estudio fue determinar la toxicidad de los piretroides lambdacialotrina, deltametrina y alfacipermetrina en L. longipalpis, a través de ensayos concentración-mortalidad. Los ensayos se hicieron siguiendo los lineamientos de la OMS, pero en lugar de los recipientes de exposición y de los papeles impregnados de la OMS, se utilizaron botellas de vidrio Wheaton de 250 ml tratadas con 1 ml de solución de insecticida en alcohol absoluto. Grupos de 10 hembras de L. longipalpis sin alimentación sanguínea fueron expuestos a cinco concentraciones de cada piretroide, que causaron entre el 5 y 100 % de mortalidad. Pasada una hora de exposición, las hembras se trasladaron a los recipientes de observación y la mortalidad se registró 24 h después. Las concentraciones (g/ml) que mataron el 50 y el 95 % (CL50 y CL95) de las hembras expuestas de L. longipalpis fueron de 0,05 y 0,86 para la lambdacialotrina, 0,24 y 3,62 para la alfacipermetrina y 0,53 y 4,72 para la deltametrina. Basados en las CL50 obtenidas, la lambdacialotrina fue el piretroide con mayor toxicidad para L. longipalpis, seguido por la alfacipermetrina y la deltametrina. Se espera que estos datos puedan ser útiles en estudios de los efectos de concentraciones sub-letales de los tres piretroides en el comportamiento de L. longipalpis y en estudios de la susceptibilidad del vector a los mismos.

Abstract Introduction: The study of the interaction between the parasite, the vector and the mammalian hosts, including man, allows to understand the behavior of the leishmaniases. Objective: To determine the presence of Lutzomyia species and to detect the Leishmania infection in Didelphis marsupialis in an endemic area for visceral leishmaniasis. Materials and methods: Phlebotomine fauna and individuals of D. marsupialis were collected with CDC and Tomahawk™ traps, respectively. The species of Lutzomyia were identified using the Young and Duncan key (1994). Ear and tail biopsies and blood samples from D. marsupialis were taken to identify the Leishmania species by amplifying a fragment of the gene associated with the 70 kD heat shock protein. Results: Seven Lutzomyia species were identified: Lu. evansi, Lu. gomezi, Lu. panamensis, Lu. dubitans, Lu. cayennensis cayennensis, Lu. rangeliana and Lu. trinidadensis. The first three species have epidemiological importance in Colombia because of their implications in the transmission of the Leishmania parasite. Sixty-five tissue samples from 19 D. marsupialis individuals were negative for Leishmania spp. Conclusions: The presence of the Lutzomyia species that have been identified as vectors for Leishmania inside and around houses in the village of El Bledo, in El Carmen de Bolívar represents a risk of infection. Furthermore, the presence of Lu. panamensis is reported for first time in El Carmen de Bolívar in Colombia. Although the lack of detection of Leishmania spp. in D. marsupialis samples may suggest that D. marsupialis does not play an important role in the transmission cycle of Leishmania in this region, it is necessary to carry out further longitudinal studies to confirm this hypothesis.


Resumen Introducción. El estudio de la interacción entre el parásito, el vector y los huéspedes mamíferos, incluido el hombre, permite entender el comportamiento de la leishmaniasis. Objetivo. Determinar la presencia de especies del género Lutzomyia y detectar la infección por Leishmania spp. en Didelphis marsupialis en un área endémica de leishmaniasis visceral. Materiales y métodos. Se recolectaron flebotomíneos y D. marsupialis con trampas CDC y Tomahawk™, respectivamente. Las especies de Lutzomyia se identificaron usando la clave de Young y Duncan, 1994. Se tomaron biopsias de oreja, cola y muestras de sangre de D. marsupialis para diagnosticar Leishmania spp. mediante la amplificación de un fragmento del gen de la proteína de choque térmico de 70 kD. Resultados. Se identificaron siete especies de Lutzomyia: Lu. evansi, Lu. gomezi, Lu. panamensis, Lu. dubitans, Lu. cayennensis cayennensis, Lu. rangeliana y Lu. trinidadensis. Las tres primeras especies son reconocidas como vectores en el país por estar implicadas en la transmisión de Leishmania spp. En total, 65 muestras de tejidos de oreja, cola y de sangre provenientes de 19 D. marsupialis fueron negativas para Leishmania spp. en la PCR-HSP70. Conclusiones. La presencia de flebotomíneos con importancia epidemiológica en la zona evaluada representa un riesgo de transmisión. Asimismo, Lu. panamensis es reportada por primera vez en El Bledo (Carmen de Bolívar). La ausencia de Leishmania spp. en D. marsupialis podría sugerir que esta especie no tiene un papel importante en el ciclo de transmisión de Leishmania en la vereda El Bledo, por lo que es necesario profundizar en estudios longitudinales para corroborar esta hipótesis.

Objective: To determine the spatial distribution and infection rate of sand flies as vectors of Leishmania parasite in Ardabil province, northwest of Iran. Methods: This was a descriptive cross-sectional study. The sand flies were collected from 30 areas in all 10 districts of Ardabil province during 2017. The specimens were caught using the sticky traps. The head and genitalia of sand flies were separated and mounted in Berlese solution for microscopic identification. The Geographical Information System ArcMap10.4.1 software was used to provide the spatial maps. Results: A total of 2794 sand flies specimens were collected and 22 species of sand flies were identified from the two genera: Phlebotomus and Sergentomyia from Ardabil province. The highest frequency was found in Phlebotomus papatasi (23.7%) followed by Phlebotomus kandelakii (13.0%). The promastigote form of Leishmania infantum parasite has been reported from the three main vectors of visceral leishmaniasis (Phlebotomus kandelakii, Phlebotomus perfiliewi and Phlebotomus tobbi) from Ardabil province, where the spatial distribution map of these visceral leishmaniasis vectors was prepared. Some important species of sand flies such as Phlebotomus kandelakii, Phlebotomus perfiliewi and Phlebotomus tobbi were reported and identified as main and probable vectors of visceral leishmaniasis in Ardabil. Conclusions: According to the Geographic Information System based maps, the frequency of the sand flies as leishmaniasis vectors, the leishmania parasite infection rate and the prevalence of the disease in the central areas of Ardabil province are higher than in other areas in Ardabil province.

Resumen Introducción. El género Warileyaes uno de los taxones menos conocidos de los flebotomíneos, el cual está integrado por solo nueve especies: Warileya (Warileya) phlebotomanica, W. (Hertigia) hertigi, W. (W.) rotundipennis, W. (W.) nigrosacculus, W. (W.) yungasi, W. (W.) fourgassiensis, W. (W.) lumbrerasi, W. (W.) euniceae y W. (W.) leponti. Objetivo. Registrar el hallazgo de una especie del género Warileya en Antioquia, Colombia. Materiales y métodos. Los flebotomíneos se recolectaron en una caverna de la Reserva Natural Cañón del Río Claro del municipio de San Francisco, departamento de Antioquia, Colombia. El muestreo entomológico sehizo con una trampa de luz de tipo CDC, durante tres noches consecutivas, en mayo de 2008. La determinación taxonómica se basó en la revisión del material tipo de la especie y de las claves estándar para flebotomíneos americanos. Resultados. Se identificaron taxonómicamente cinco machos y dos hembras de flebotomíneos comoW. (H.) hertigi. En ambos sexos fue notoria la ausencia de cicatrices de setas en el anepisternón, el proepimerón y el clípeo, la presencia de dos hileras transversales de cicatrices de setas en los tergitos, y la longitud corta de la vena gamma. Conclusión. El hallazgo de W. (H.) hertigi incrementa a 64 el número de especies de flebotomíneos encontradas en el departamento de Antioquia. En total, 164 especies de flebotomíneos han sido registradas en Colombia.


Abstract Introduction: The genus Warileyais one of the least-known taxa of sandflies, comprising only nine species, i.e., W. (Warileya) phlebotomanica, W. (Hertigia) hertigi, W. (W.) rotundipennis, W. (W.) nigrosacculus, W. (W.) yungasi, W. (W.) fourgassiensis, W. (W.) lumbrerasi, W. (W.) euniceae and W. (W.) leponti. Objective: To document the presence of a species of the genus Warileya in Antioquia, Colombia. Materials and methods: Sandflies were collected in a cavern of the Cañóndel Río Claro Natural Reserve, of the municipality of San Francisco, Antioquia department, Colombia. Phlebotomine sampling was carried out using a CDC light trap during three consecutive nights in May of 2008. Taxonomical determination was based on a revision of the type material of the species and through the use of standard keys for American sandflies. Results: Five male and two female sandflies were taxonomically identified as W. (H.) hertigi. In both sexes, the absence of setal scars in the anepisternum, proepimeron and clypeus; the presence of two transverse rows of setal scars in the tergites; and the short length of the vena gamma were notable. Conclusion: The finding of W. (H.) hertigi increases the number of sandfly species found in Antioquia department to 64. In total, 164 sandfly species have been recorded in Colombia.

Resumen Introducción: Nyssomyia yuilli (Young & Porter, 1972) y Nyssomyia trapidoi (Farchild & Hertig, 1952) son vectores de Leishmania panamensis (Lainson y Shaw, 1972), causal de leishmaniasis cutánea en varios países de América como Brasil, Colombia, Ecuador y Panamá. Objetivo: Describir la actividad nocturna de N. yuilli y N. trapidoi en el municipio de Otanche, zona endémica de leishmaniasis cutánea. Metodología: Los especímenes fueron colectadas por medio de trampas CDC localizadas en el intra y peri-domicilio de viviendas que tenían antecedentes o casos activos de personas infectadas con leishmaniasis cutánea. Resultados: N. yuilli presentó una mayor actividad cerca de la media noche, mientras que N. trapidoi no mostró ningún incremento y mantuvo una actividad constante durante toda la noche. Conclusiones: Se observó que N. yuilli tiene una conducta típica de una especie de bosque con picos de actividad a media noche, mientras que N. trapidoi tiene un comportamiento de una especie antropofílica que se ha adaptado a la vida cerca de las viviendas humanas, con una actividad constante durante toda la noche sin ningún aumento esporádico.


Abstract Introduction: Nyssomyia yuilli (Young & Porter, 1972) and Nyssomyia trapidoi (Farchild & Hertig, 1952) are vectors of Leishmania panamensis (Lainson y Shaw, 1972), causal of cutaneous leishmaniasis in several American countries such as Brazil, Colombia, Ecuador and Panama. Objective: Describe the nocturnal activity of N. yuilli and N. trapidoi in Otanche Municipality, endemic zone of cutaneous leishmaniasis. Methods: The specimens were collected through CDC traps located in the intra and peri-domicile of homes that had antecedents or active cases of infected people with cutaneous leishmaniasis. Results: N. yuilli present increased activity near midnight, while N. trapidoi not show any increase and maintained a constant activity throughout the night. Conclusions: It was observed that N. yuilli demonstrates typical conduct of a forest species, with peaks of activity at midnight, while N. trapidoi shows behaviour of an anthropophilic species that has been adapted to life near to human residences, with constant activity throughout the night without any sporadic increases.

INTRODUÇÃO: A leishmaniose Visceral (LV) é uma antropozoonose causada pela Leishmania infantum que avança nos últimos trinta anos a passos largos em sentido ao ambiente urbano. Desde 1977 há registros da doença em áreas periféricas do município do Rio de Janeiro, entretanto em 2011 foi descrita no bairro do Caju por meio do diagnóstico do parasita em cães infectados e da captura de espécimes de seu principal vetor. OBJETIVO: Este artigo tem por objetivo divulgar o trabalho primário de captura e identificação entomológica destes espécimes que proporcionou ações de vigilância no primeiro foco ativo urbano de LV na cidade do Rio de Janeiro, citado recentemente em diversos estudos, mas ainda não publicizado em todos os seus detalhes. MÉTODOS: As armadilhas luminosas CDC adaptadas foram instaladas de forma setorizada em área circular de 1000 m de raio a partir do suposto ponto focal, das 17h00m às 07h00m da manhã seguinte, entre junho e setembro de 2011. RESULTADOS: Foram capturados 253 espécimes de Lutzomyia longipalpis, dos quais 63,1% na área do canil, considerado o foco primário de dispersão dos vetores. CONCLUSÃO: Os resultados descritos corroboram com artigos que indicam a urbanização do principal vetor da LV, salientando a importância de estudos mais detalhados sobre o seu ciclo biológico urbano e a necessidade da mudança dos paradigmas da vigilância das leishmanioses no Brasil.


INTRODUCTION: Leishmaniasis, Visceral (LV) is an anthropozoonosis caused by Leishmania infantum that was advanced in the last thirty years in the urban environment. Since 1977 there are records of the disease in peripheral areas of the municipality of Rio de Janeiro, however, in 2011 it was described in the Caju neighborhood by means of the diagnosis of the parasite in infected dogs and the capture of specimens of its main vector. OBJECTIVE: This article aims to disclose the primary work of entomological capture and identification of these specimens that provided surveillance actions in the first urban active focus of LV in the city of Rio de Janeiro, quoted recently in several studies, but not publicized in all its details. METHODS: The adapted CDC light traps were installed in a circular sector of 1000 m radius from the supposed focal point, from 5:00 p.m. to 7:00 p.m. the following morning, between June and September 2011. RESULTS: A total of 253 specimens of Lutzomyia longipalpis were collected, 63.1% of which in the kennel area, considered the primary focus of dispersion of the vectors. CONCLUSION: The results described corroborate articles indicating the urbanization of the main vector of LV, stressing the importance of more detailed studies of its urban life cycle and the need of the changing paradigms of leishmaniasis surveillance in Brazil.

Resumen Introducción. Las técnicas de biología molecular han permitido ampliar el conocimiento sobre las fuentes de ingestión de sangre de los insectos vectores. Sin embargo, la utilidad de estas técnicas depende de la cantidad de sangre ingerida y del proceso de digestión en el insecto. Objetivo. Determinar el tiempo límite de detección del gen citocromo b (Cyt b) de humanos en hembras de Lutzomyia evansi alimentadas experimentalmente. Materiales y métodos. Se evaluaron ocho grupos de hembras de L. evansi alimentadas con sangre humana, las cuales fueron sacrificadas en intervalos de 24 horas desde el momento de la ingestión sanguínea. Se extrajo el ADN total de cada hembra y se amplificó un segmento de 358 pb del gen Cyt b. Los productos amplificados fueron sometidos a un análisis de polimorfismos en la longitud de los fragmentos de restricción (Restriction Fragment Length Polymorphism, RFLP), con el fin de descartar falsos positivos. Resultados. El segmento del gen Cyt b de humanos fue detectado en 86 % (49/57) de las hembras de L. evansi a partir de las 0 horas y hasta 168 horas después de la ingestión de sangre. En 7 % (4/57) de los individuos se amplificó el ADN del insecto y en el 7 % restante no se amplificó la banda de interés. No se encontraron diferencias estadísticas en cuanto a la amplificación del segmento del gen Cyt b de humanos ni al número de muestras amplificadas entre los grupos de hembras sacrificadas a distintas horas después de la ingestión. Conclusión. El segmento del gen Cyt b de humanos fue detectable en hembras de L. evansi hasta 168 horas después de la ingestión de sangre.


Abstract Introduction: Molecular biology techniques have allowed a better knowledge of sources of blood meals in vector insects. However, the usefulness of these techniques depends on both the quantity of ingested blood and the digestion process in the insect. Objective: To identify the time limit for detection of the human cytochrome b (Cyt b) gene in experimentally fed females of Lutzomyia evansi. Materials and methods: Eight groups of L. evansi females were fed on human blood and sacrificed at intervals of 24 hours post-ingestion. Total DNA was extracted from each female and a segment of 358 bp of Cyt b was amplified. In order to eliminate false positives, amplification products were subjected to a restriction fragment length polymorphism (RFLP) analysis. Results: The human Cyt b gene segment was detected in 86% (49/57) of the females of L. evansi, from 0 to 168 hours after blood ingestion. In 7% (4/57) of the individuals we amplified insect DNA, while in the remaining 7%, the band of interest was not amplified. We did not find any statistical differences between groups of females sacrificed at different times post-blood meal regarding the amplification of the human Cyt b gene segment or the number of samples amplified. Conclusion: The human Cyt b gene segment was detectable in L. evansi females up to 168 hours after blood ingestion.

Resumen Introducción. Los departamentos de Chocó y Antioquia en Colombia presentan condiciones climáticas y de vegetación que favorecen el establecimiento de especies de vectores del género Lutzomyia y la transmisión de Leishmania spp. a poblaciones humanas que ingresan a ambientes selváticos conservados. Objetivo. Reportar las especies de flebotomíneos presentes en tres reservas naturales de las regiones del Darién y del Pacífico en Colombia. Materiales y métodos. Los flebotomíneos se recolectaron en las reservas naturales El Aguacate (Acandí, Chocó), Nabugá (Bahía Solano, Chocó) y Tulenapa (Carepa, Antioquia). La recolección se hizo con trampas de luz CDC, mediante búsqueda activa en sitios de reposo y con trampas Shannon. La determinación taxonómica de especies se basó en las claves taxonómicas. En algunas especies de interés taxonómico, se evaluó el uso de secuencias parciales de la región 5' del gen COI. Resultados. Se recolectaron 611 flebotomíneos adultos: 531 en Acandí, 45 en Carepa y 35 en Bahía Solano. Se identificaron 17 especies del género Lutzomyia, tres del género Brumptomyia y una del género Warileya. Las distancias genéticas (K2P) y los soportes de agrupación (>99 %) en el dendrograma de neighbor joining correspondieron a la mayoría de unidades taxonómicas operacionales moleculares (Molecular Operational Taxonomic Units, MOTU) establecidas para el grupo Aragaoi y confirmaron claramente la identidad de Lu. coutinhoi. Conclusión. Se identificaron especies que tienen importancia en la transmisión de la leishmaniasis en Acandí, Bahía Solano y Carepa. Se confirmó la presencia de Lu. coutinhoi en Colombia.


Abstract Introduction: The departments of Chocó and Antioquia in Colombia show climatic and vegetation conditions favoring the establishment of vector species of the genus Lutzomyia and the transmission of Leishmania spp. to human populations entering conserved forest environments. Objective: To report the species of Phlebotomine sandflies present in three natural reserves in the Darien and Pacific regions of Colombia. Materials and methods: Sand flies were collected specifically in the natural reserves El Aguacate (Acandí, Chocó), Nabugá (Bahía Solano, Chocó) and Tulenapa (Carepa, Antioquia). Sand flies were collected with CDC light traps, active search in resting places and Shannon traps. The taxonomic determination of species was based on taxonomic keys. For some species of taxonomic interest, we evaluated the partial sequences of the 5' region of COI gene. Results: A total of 611 adult sand flies were collected: 531 in Acandí, 45 in Carepa and 35 in Bahía Solano. Seventeen species of the genus Lutzomyia, three of the genus Brumptomyia and one of the genus Warileya were identified. The genetic distances (K2P) and grouping supported (>99%) in the neighbor joining dendrogram were consistent for most established molecular operational taxonomic units (MOTU) of the Aragaoi group and clearly confirmed the identity of Lu. coutinhoi. Conclusion: Species that have importance in the transmission of leishmaniasis in Acandí, Bahía Solano and Carepa were identified. The presence of Lu. coutinhoi was confirmed and consolidated in Colombia.

Resumen Introducción. La provincia de Pichincha, Ecuador, es un área endémica de leishmaniasis cutánea, en donde se han determinado como vectores los flebotomíneos antropofílicos con infección natural por Leishmania spp. Sin embargo, no se ha evaluado el papel en la transmisión de las especies zoofílicas. Objetivo. Evaluar la infección natural por Leishmania en dos especies de flebotomíneos zoofílicos, Lutzomyia reburra y Lu. barrettoi majuscula, y en una antropofílica, Lu. trapidoi, así como la endofagia y la sinantropía de estas especies en el noroccidente de Pichincha. Materiales y métodos. Los flebotomíneos se recolectaron en trampas de luz CDC colocadas en diferentes hábitats y altitudes en sitios que son focos de leishmaniasis cutánea. La infección con Leishmania spp. se detectó en el ADN genómico de hembras de las especies de flebotomíneos de interés. Se amplificó el gen espaciador interno de la transcripción del ARN ribosómico, unidad I (ITS1), y los genes de las topoisomerasas mitocondrial II (mtTOPOII) y nuclear II (TopoII). Se determinaron los porcentajes de positividad para Leishmania a escala espaciotemporal, la proporción de endofagia y el índice de sinantropía. Resultados. Se determinó la presencia de infección natural por Le. amazonensis en Lu. reburra (9,5 %) y Lu. b. majuscula (23,8 %); en Lu. trapidoi se detectaron Le. amazonensis, Le. brazilienis y Le. naiffilainsoni. Los flebotomíneos eran asinantrópicos y con baja endofagia. Conclusión. Se registró por primera vez la presencia de infección natural en Lu. reburra y Lu. barrettoi majuscula por Le. amazonensis, y se demostró la importancia de los flebotomíneos zoofílicos en el mantenimiento del ciclo de transmisión de Leishmania spp. en focos endémicos.


Abstract Introduction: The province of Pichincha in Ecuador is an endemic area of cutaneous leishmaniasis, where anthropophilic sand flies with natural infection by Leishmania, have been reported as vectors. However, the role in transmission of zoophilic species has not been evaluated. Objective: To evaluate natural infection by Leishmania in two zoophilic phlebotomine sand fly species, Lutzomyia reburra and Lu. barrettoi majuscula, and one anthropophilic species, Lu. trapidoi, as well as the endophagy and synanthropism of these species in the northwest of Pichincha. Materials and methods: Phlebotomines were collected using CDC light traps in different habitats and altitudes with presence of cutaneous leishmaniasis. Leishmania infection was detected using genomic DNA from females of the collected sand flies. We amplified the internal transcribed spacer gene of ribosomal RNA I (ITS1), the mitochondrial topoisomerase II gene (mtTOPOII), and the nuclear topoisomerase II gene (TopoII). Percentages of positivity for Leishmania, at spatio-temporal scale, proportion of endophagy and synanthropism index were calculated. Results: Natural infection was determined for Le. amazonensis in Lu. reburra (9.5%) and Lu. b. majuscula (23.8%), while in Lu. trapidoi we detected Le. amazonensis, Le. brazilienis and Le. naiffilainsoni. Phlebotomines were asynanthropic and with low endophagy. Conclusion: Natural infection with Le. amazonensis was recorded for the first time in Lu. reburra and Lu. b. majuscula, demonstrating the importance of zoophilic phlebotomines in the maintenance of the Leishmania transmission cycle in endemic foci.

BACKGROUND Maxadilan (Max) is a salivary component in the sandfly Lutzomyia longipalpis (Lutz & Neiva 1912), a vector of visceral leishmaniasis. Max has a powerful vasodilatory effect and is a candidate vaccine that has been tested in experimental leishmaniasis. Nyssomyia neivai (Pinto 1926) is a vector of the pathogen responsible for American tegumentary leishmaniasis (ATL) in Brazil. OBJECTIVE We searched for Max expression in Ny. neivai and for antibodies against Max in ATL patients. METHODS cDNA and protein were extracted from the cephalic segment, including salivary glands, of Ny. neivai and analysed by polymerase chain reaction, DNA sequencing, and blotting assays. The results were compared with data obtained from Lu. longipalpis samples. We quantified antibodies against Max in serum samples from 41 patients with ATL (31 and 10 with the cutaneous and mucocutaneous forms, respectively) and 63 controls from the endemic northeastern region of São Paulo state, using enzyme-linked immunosorbent assay. FINDINGS Recognition of a Max-simile peptide by specific antibodies confirmed expression of a Max sequence in Ny. neivai (GenBank EF601123.1). Compared to controls, patients with ATL presented higher levels of antibodies against Max (p = 0.004); 24.4% of the patients with ATL and 3.2% of the controls presented anti-Max levels above the cutoff index (p = 0.014). The anti-Max levels were not associated with the specific clinical form of ATL, leishmanin skin test response, absence or presence of amastigotes in histopathologic exam, results of indirect immunofluorescence testing for leishmaniasis, or duration of cutaneous form disease. MAIN CONCLUSION High serum anti-Max levels did not protect patients against ATL, but confirmed previous natural exposure to Ny. neivai bites in this ATL endemic region.

ABSTRACT

Canine visceral leishmaniasis is a zoonotic disease caused by parasites of the genus Leishmania and transmitted by phlebotomine sandflies of the genus Lutzomyia in the New World and Phlebotomus in the Old World. The individual protection of dogs by treatment with permethrin 65% or by a 4% deltamethrin-impregnated collar is evaluated in this study based on the antifeeding of the products and the short term insecticidal antifeeding effects in the following sites on the dog's body: the inner side of the ear and the inguinal region. Both products produced a decrease in blood feeding and an increase in the antifeeding of the sandflies and the effects were not homogeneous, as there were significant differences between the effects on the pinna and on the inguinal region. The treatments of dogs with synthetic pyrethroids are important strategies to reduce the infection rates in endemic regions however the antifeeding and antifeeding effects were low and may not be sufficient to ensure a dog's individual protection.


Leishmaniose visceral canina é uma zoonose causada por parasitos do gênero Leishmania e transmitida por flebótomos do gênero Lutzomyia no Novo Mundo e Phlebotomus no Velho Mundo. A proteção individual do cão através do tratamento com produtos contendo permetrina 65% ou com a coleira impregnada com deltametrina 4% é avaliada no presente estudo, baseado nos efeitos de não alimetação e inseticida imediatos no pavilhão auditivo direito e região inguinal direita. Ambos os produtos demonstraram redução no ingurgitamento das fêmeas e aumento da mortalidade, mas o efeito não foi homogêneo, uma vez que foram verificadas diferenças estatisticamente significativas nos efeitos nas diferentes regiões. O tratamento dos cães com piretroides é uma importante estratégia para redução das taxas de infecção em regiões endêmicas, no entanto, os efeitos mortalidade e repelência foram baixos e podem não ser suficientes para a proteção individual do cão.

Los insectos relacionados con la transmisión de los patógenos causantes de las leishmaniasis han sido poco estudiados en la Sierra Nevada de Santa Marta, Colombia, incluido el departamento de Magdalena, donde a la fecha están registradas trece especies del género Lutzomyia. En la presente nota se informa el hallazgo de tres especies y un subgénero adicionales en la región. Se recolectaron 885 flebotomíneos en Seywiaka y las veredas Las Tinajas y Calabazo, estribaciones de la Sierra Nevada de Santa Marta (117-130 m s.n.m.). El 84 % de los ejemplares se obtuvieron con trampa CDC, el 11 % con trampa Shannon y el 5 % fueron capturados, en reposo, con un dispositivo eléctrico de succión. Se identificaron nueve especies, Lu. gomezi, Lu. panamensis, Lu. trinidadensis, Lu. carpenteri, Lu. evansi, Lu. dysponeta, Lu. dubitans, Lu. shannoni, y Lu. micropyga, la más abundante fue Lu. gomezi (69 %), seguida por Lu. panamensis (14 %). También se recolectaron ejemplares de la serie Lu. osornoi del subgénero Helcocyrtomyia. Entre el material hallado sobresalen Lu. carpenteri, Lu. dubitans y Lu. dysponeta como primeros registros para el departamento del Magdalena, además de Lu. (Helcocyrtomyia) sp., que representa el primer informe del subgénero en el Caribe colombiano.


Phlebotomine sand flies, vectors of leishmaniasis, have not been well studied in the Sierra Nevada de Santa Marta, and likewise, are not well known in other regions of the Department of Magdalena, Colombia. To date only thirteen species of Lutzomyia have been recorded as occurring in the Department. The present note adds three species and includes an additional subgenus. Collections were made in the lower foothills of the Sierra Nevada de Santa Marta at elevations ranging from 117-130 m in the communities of Seywiaka, Las Tinajas and Calabazo. Eighty-four percent of the 885 phlebotomines sand flies collected were obtained from CDC light traps, 11 % from Shannon trap and 5 % from typical resting sites using an electric aspirator. The following nine species were identified from the collections: Lutzomyia gomezi, Lu. panamensis, Lu. trinidadensis, Lu. carpenteri, Lu. evansi, Lu. dysponeta, Lu. dubitans, Lu. shannoni, and Lu. micropyga. The most abundant species were Lu. gomezi and Lu. panamensis, which, respectively, accounted for 69 % and 14 % of the specimens. Of the nine species, Lu. carpenteri, Lu. dubitans and Lu. dysponeta represent new records for the Department of Magdalena. Also, a few female specimens were encountered of a species belonging to the Lu. osornoi series of the subgenus Helcocyrtomyia, which represents the first record of this subgenus in the Caribbean region of Colombia.

Este estudo foi realizado em Belo Horizonte (Minas Gerais, Brasil), área endêmica para Leishmaniose visceral (LV), onde a doença vem se expandindo rapidamente, com 1.612 casos registrados nos últimos 10 anos. Nossa pesquisa avaliou o perfil epidemiológico em duas Áreas de abrangência (AA) dos Centros de Saúde (CS) Miramar e Salgado Filho do município de Belo Horizonte, com transmissão recente de LV, buscando compreender a tríade envolvida na transmissão: parasito-vetor-reservatório. Cada AA foi dividida em três trechos, controle, borrifação e manejo, a fim de avaliar o impacto das medidas de controle no número mensal de Lutzomyia longipalpis, através da curva flebotomínica. Em todos os trechos a captura de flebotomíneos e o inquérito canino censitário foram realizados.Durante 24 meses (setembro/2010 a agosto/2012) foram capturados 5194 flebotomíneos, distribuídos em 6 espécies, sendo Lu. longipalpis, vetora da LV, a espécie predominante (96,54%). Houve uma correlação entre a precipitação pluviométrica e o número de Lu. longipalpis capturados, mostrando que o número de espécimes de flebotomíneos capturados aumentou após o período chuvoso...


Através do uso da Leishmania Nested PCR e sequenciamento, foram analisados 93 “pools” espécie-específicos. Leishmania infantum e Le. braziliensis foram encontradas em 23 e em 2 dos 26 “pools” de Lu. longipalpis, respectivamente. O DNA de Le. infantum foi encontrado em um “pool” de cada uma das espécies Lu. lloydi e complexo cortelezzii. A taxa mínima de infecção natural de flebotomíneos na área foi alta, 18,8% para Lu. Longipalpis, 20% para complexo cortelezzii e 100% para Lu. lloydi. Em relação às medidas de controle, observamos uma redução estatisticamente significativa no número de flebotomíneos no trecho manejo ambiental, enquanto houve apenas uma tendência de queda no número de flebotomíneos no trecho borrifação. Em relação ao reservatório doméstico, um total de 1408 cães foram examinados em 2011 através de testes sorológicos; 3,6% apresentaram-se soropositivos para LV canina (LVC). Biópsias de linfonodo, pele, baço e aspirado de medula óssea destes animais foram obtidos e analisados através do diagnóstico parasitológico e molecular. A positividade tanto para mielocultura como para o técnica de aposição em lâmina foi de 72.5%. Todas as amostras de baço mostraram-se positivas para LnPCR, indicando a presença de Le. infantum nas 51 amostras. O linfonodo foi o tecido menos sensível na PCR (57%) em relação aos demais. Cães sintomáticos demonstraram uma tendência a ter mais testes positivos. Nossos resultados mostram que as interfaces parasito-vetor e parasito-reservatório são ativas em áreas de médio risco para transmissão de LV, e que ações eficazes e rápidas devem ser implementadas para controlar a expansão da doença...

Se presenta una lista actualizada de especies de flebotomíneos en zonas cafeteras de la región andina colombiana. Con base en la revisión y verificación taxonómica, se registraron 53 especies presentes en 12 departamentos. Además de los registros obtenidos con base en un muestreo intensivo en cinco departamentos, se recopilaron los datos existentes en trabajos publicados y en la revision taxonómica de los especímenes de la zona pertenecientes a la colección entomológica del Programa de Estudio y Control de Enfermedades Tropicales (PECET). El listado comprende los géneros Brumptomyia (2 especies), Lutzomyia (50 especies) y Warileya (1 especie). Con base en este trabajo se confirmaron 11 nuevos registros de especies en la región cafetera colombiana, entre los cuales es relevante Lutzomyia panamensis , especie de importancia médica no registrada previamente en esta zona. En total, 18 especies de las registradas poseen hábitos antropofílicos o están relacionadas con la transmisión de Leishmania spp.


An updated list of phlebotomine sand flies species in coffee growing areas in the Colombian Andean region is presented. Fifty three species were reported from 12 departments. In addition, species distribution in the region was derived from specimens obtained during intensive field work in five departments, from previously published studies and from the taxonomic revision of specimens in the entomological collection of the Programa de Estudio y Control de Enfermedades Tropicales (PECET). The list includes the genera Brumptomyia (2 species), Lutzomyia (50 species) and Warileya (1 species). The updated list contains eleven new records in the region under study, including Lutzomyia panamensis , a species of medical importance not recorded previously in this zone. Eighteen of the species are considered to be anthropophilic, and many of them have been implicated in the transmission of leishmaniasis.
